Dr. Mario on Tetriksen kaltainen pulmapeli, jossa Nintendon maskotti Mario toimii lääkärinä. Pelissä pudotetaan kaksipuolisia vitamiinikapseleita pelikentälle, joka on 8 ruutua leveä ja 16 ruutua korkea ja muistuttaa lääkepulloa. Kentällä on kolmenvärisiä viruksia.
